Adjusting Leading in Illustrator

To space lines of text by hand, use the leading meter on the character palette. Leading controls how the lines of text are spaced. Open the character palette by going to Window > Type > Character in Illustrator 10, or Type > Character in Illustrator 9, or type command/control +T.

The text in this example is 12 pt Myriad, and the leading is set at 14.5, which is what you get if you choose Auto in the leading dropdown.

To adjust it, use the up and down arrows on the leading control to make the space greater or move the text closer together.

This is the same text but the leading was changed to 21 pt. to move the text lines farther apart.
